    Molecular survey of tick-borne pathogens in Ixodid ticks collected from hunted wild animals in Tuscany, Italy

    Objective: The aim of the present study was to determine the prevalence of zoonotic tick-borne bacteria in feeding ticks removed from hunted wild animals. Methods: PCR was executed on DNA extracted from 77 tick pools to detect Anaplasma phagocytophilum, Bartonella spp., Borrelia burgdorferi sensu lato, Coxiella burnetii and Rickettsia spp. Results: A total of 432 ticks were collected: 30 (6.94%) Haemaphysalis punctata, 72 (16.7%) Dermacentor marginatus and 330 (76.38%) Ixodes ricinus. For each animal one or two pools of 3 ticks of the same species was constituted. Seventy-seven tick pools were examined by PCR: 58 (75.32%) resulted infected and among them 14 (18.18%) showed co-infections. In particular, 29 (37.66%) pools were positive for Bartonella spp., 23 (29.87%) for A. phagocytophilum, 16 (20.78%) for Rickettsia spp., and 5 (6.49%) for B. burgdorferi s.l. All samples were negative for C. burnetii. Conclusion: The results demonstrate the presence of several zoonotic tick-borne pathogens in the studied area, and underline the risk of exposure to infections for hunters not only during the outdoor activity, but also when they manipulate hunted animals infested by infected ticks

    From September 2009 to February 2011, 122 coypus (Myocastor coypus) and 74 rats (Rattus norvegicus) were captured employing cage traps in a protected wetland in Tuscany (central Italy). Blood serum samples were collected from the animals and successively examined by the microagglutination test for several serovars of Leptospira: Bratislava, Ballum, Bataviae, Grippothyphosa, Icterohaemorrhagiae, Copenhageni, Mini, Pomona, Zanoni, Sejroe, Hardjo and Tarassovi. Kidney samples were collected from each animal and tested by bacteriological methods and submitted to Polymerase Chain Reaction. Thirty-four (27.87 %) coypu sera were positive to Leptospira interrogans serovar Bratislava, with antibody titers ranging from 1:100 to 1:400; no strain was isolated from coypu by bacteriological examination, while 12 (9.83 %) subjects were positive to PCR. All rats resulted seronegative; thirty-seven (50 %) Leptospira strains were isolated from rat kidneys; 30 were classified as Leptospira interrogans serovar Icterohaemorrhagiae and 7 as Leptospira interrogans serovar Ballum by the cross-agglutination test. Forty-five (60.81 %) rats resulted positive to PCR: 37 subjects positive to bacteriological examination and there were eight from which no strain were isolated from kidneys. These results would seem to suggest the minor zooepidemiological role of coypu in leptospirosis, which is widespread according to literature.Od rujna 2009. do veljače 2011. u zamke su bile uhvaćene 122 nutrije (Myocastor coypus) i 74 štakora selca (Rattus norvegicus) u zaštićenoj močvari u Toskani u središnjoj Italiji. Prikupljeni uzorci krvnog seruma bili su pretraženi mikroaglutinacijskim testom na nekoliko serovarova leptospira: Bratislava, Ballum, Bataviae, Grippothyphosa, Icterohaemorrhagiae, Copenhageni, Mini, Pomona, Zanoni, Sejroe, Hardjo i Tarassovi. Uzorci tkiva bubrega, uzeti od svake životinje, bili su pretraženi bakteriološki i lančanom reakcijom polimerazom. Tridesetčetiri (27,87 %) uzorka seruma nutrije bila su pozitivna na vrstu Leptospira interrogans serovar Bratislava s titrom protutijela od 1:100 do 1:400. Bakteriološkom pretragom iz nutrija nije bio izdvojen nijedan izolat, dok je 12 uzoraka (9,83 %) bilo pozitivno PCR-om. Svi štakori bili su serološki negativni, ali je 37 (50 %) izolata leptospira bilo izdvojeno iz tkiva njihovih bubrega. Od toga je 30 izolata pripadalo vrsti Leptospira interrogans serovaru Icterohaemorrhagiae, a 7 serovaru Ballum pretragom križnim aglutinacijskim testom. Ukupno su 45 štakora (60,81 %) bila pozitivna pretragom PCR-om, a 37 bakteriološkom pretragom. Od osam štakora nije bio izdvojen nijedan izolat leptospira iz bubrega. Ti rezultati upućuju na neznatnu zooepidemiološku ulogu nutrija u širenju leptospiroze

    Design and experimental tests of active control barriers for low-frequency stationary noise reduction in urban outdoor environment

    Active noise control (ANC) techniques are based on the emission of an antiphase signal in order to cancel the noise produced by a primary source. ANC has been successfully applied especially for reducing noise in confined environments, such as headphones and ducts. In this study, we present an application of ANC concepts to the design of an anti-noise barrier for an outdoor environment and its experimental testing. Even though passive techniques are effective in noise reduction at middle-high frequencies, they become less efficient at low frequencies (below 300 Hz) due to the limited dimensions of commonly deployable barriers. In this paper, we analyze the properties of a low-cost active noise system able to efficiently operate on stationary, almost pure-tone, low-frequency noise, such as that produced by electrical transformers and reactors in power and transformation plants. A prototype has been implemented and on-the-field experimental tests have been carried out. The results (confirmed also by numerical simulations) demonstrate a remarkable efficiency in the far field, with a reduction up to 15 dB with respect to the absence of the ANC system

    Pathotypes and Antimicrobial Susceptibility of Escherichia Coli Isolated from Wild Boar (Sus scrofa) in Tuscany

    Wild boar are among the most widespread wild mammals in Europe. Although this species can act as a reservoir for different pathogens, data about its role as a carrier of pathogenic and antimicrobial-resistant Escherichia coli are still scarce. The aim of this study was to evaluate the occurrence of antimicrobial-resistant and pathogenic Escherichia coli in wild boar in the Tuscany region of Italy. During the hunting season of 2018-2019, E. coli was isolated from 175 of 200 animals and subjected to antimicrobial resistance tests and PCR for detection of resistance and virulence factor genes. The highest resistance rates were against cephalothin (94.3%), amoxicillin-clavulanic acid (87.4%), ampicillin (68.6%), and tetracycline (44.6%). The most detected resistance genes were blaCMY-2 (54.3%), sul1 (38.9%), sul2 (30.9%), and tetG (24.6%). Concerning genes encoding virulence factors, 55 of 175 isolates (31.4%) were negative for all tested genes. The most detected genes were hlyA (47.4%), astA (29.1%), stx2 (24.6%), eaeA (17.1%), and stx1 (11.4%). E. coli was classified as Shiga toxin-producing E. coli (STEC) (21.7%), enterohemorrhagic E. coli (EHEC) (6.3%), enteroaggregative E. coli (EAEC) (5.1%), and atypical enteropathogenic E. coli (aEPEC) (3.4%). Enterotoxigenic E. coli (ETEC), enteroinvasive E. coli (EIEC), and typical enteropathogenic E. coli (tEPEC) were not detected. Our results show that wild boars could carry pathogenic and antimicrobial-resistant E. coli, representing a possible reservoir of domestic animal and human pathogens

    EFFETTO DELL’ADDIZIONE DI POLLINE D’API BIOLOGICO ESSICCATO SULLO SVILUPPO DI MICRORGANISMI PATOGENI IN LATTE

    Lo scopo del presente lavoro è stato quello di valutare l'effetto dell'addizione di una sospensione di polline d'api biologico essiccato sullo sviluppo in latte di alcuni microrganismi patogeni. Il polline utilizzato è stato raccolto da api allevate in provincia di Lucca (Toscana, Italia) nell’anno di vegetazione 2013 ed è stato utilizzato per preparare una sospensione da addizionare in ragione dell’ 1%, 2%, 4% (v/v) a Skim Milk inoculato con ceppi patogeni di referenza in cariche pari a 106 UFC/mL. I ceppi indagati sono stati Enterococcus faecalis ATCC 19433, Staphylococcus aureus ATCC 6539, Listeria monocytogenes ATCC 7644, Salmonella enterica sv. Thyphimurium ATCC 14028. Lo sviluppo dei microrganismi è stato monitorato dopo 24 e 48 ore dall’inoculo in campioni mantenuti a temperatura di refrigerazione e a temperatura ottimale di crescita. E’ stato inoltre valutato il profilo microbiologico del polline impiegato. Il polline utilizzato presentava una carica batterica mesofila pari a 4,9 log UFC/g, con cariche in Enterobacteriaceae e Lactobacillus spp. pari a 3,7 log UFC/g e 4,9 log UFC/g, rispettivamente. Muffe e lieviti presentavano una carica di 5,5 log UFC/g. Variazioni nelle conte in latte sono state rilevate solamente dopo incubazione a temperatura ottimale di sviluppo. A 48 h di incubazione E. faecalis in latte al 4% di polline ha mostrato una diminuzione delle cariche pari a 1 log UFC/mL; S. aureus è diminuito di 2 log ufc/ml in latte al 2% e 4% di polline; L. monocytogenes è diminuita di quasi 4 log UFC/mL, sempre in latte al 4% di polline. Infine, S. enterica ha mostrato ben 6 log UFC/mL di diminuzione dopo 48 ore in latte al 4% di polline. Analisi ulteriori sono necessarie al fine di valutare in quale entità le diminuzioni riscontrate siano dovute ai composti antimicrobici presenti nel polline o piuttosto ad un effetto di competizione biologica esercitato dalla microflora naturalmente presente nel polline

    Identification of evolutionarily conserved genetic regulators of cellular aging

    To identify new genetic regulators of cellular aging and senescence, we performed genome-wide comparative RNA profiling with selected human cellular model systems, reflecting replicative senescence, stress-induced premature senescence, and distinct other forms of cellular aging. Gene expression profiles were measured, analyzed, and entered into a newly generated database referred to as the GiSAO database. Bioinformatic analysis revealed a set of new candidate genes, conserved across the majority of the cellular aging models, which were so far not associated with cellular aging, and highlighted several new pathways that potentially play a role in cellular aging. Several candidate genes obtained through this analysis have been confirmed by functional experiments, thereby validating the experimental approach. The effect of genetic deletion on chronological lifespan in yeast was assessed for 93 genes where (i) functional homologues were found in the yeast genome and (ii) the deletion strain was viable. We identified several genes whose deletion led to significant changes of chronological lifespan in yeast, featuring both lifespan shortening and lifespan extension. In conclusion, an unbiased screen across species uncovered several so far unrecognized molecular pathways for cellular aging that are conserved in evolution

    Acute Delta Hepatitis in Italy spanning three decades (1991–2019): Evidence for the effectiveness of the hepatitis B vaccination campaign

    Updated incidence data of acute Delta virus hepatitis (HDV) are lacking worldwide. Our aim was to evaluate incidence of and risk factors for acute HDV in Italy after the introduction of the compulsory vaccination against hepatitis B virus (HBV) in 1991. Data were obtained from the National Surveillance System of acute viral hepatitis (SEIEVA). Independent predictors of HDV were assessed by logistic-regression analysis. The incidence of acute HDV per 1-million population declined from 3.2 cases in 1987 to 0.04 in 2019, parallel to that of acute HBV per 100,000 from 10.0 to 0.39 cases during the same period. The median age of cases increased from 27 years in the decade 1991-1999 to 44 years in the decade 2010-2019 (p < .001). Over the same period, the male/female ratio decreased from 3.8 to 2.1, the proportion of coinfections increased from 55% to 75% (p = .003) and that of HBsAg positive acute hepatitis tested for by IgM anti-HDV linearly decreased from 50.1% to 34.1% (p < .001). People born abroad accounted for 24.6% of cases in 2004-2010 and 32.1% in 2011-2019. In the period 2010-2019, risky sexual behaviour (O.R. 4.2; 95%CI: 1.4-12.8) was the sole independent predictor of acute HDV; conversely intravenous drug use was no longer associated (O.R. 1.25; 95%CI: 0.15-10.22) with this. In conclusion, HBV vaccination was an effective measure to control acute HDV. Intravenous drug use is no longer an efficient mode of HDV spread. Testing for IgM-anti HDV is a grey area requiring alert. Acute HDV in foreigners should be monitored in the years to come
